ASK1

Apoptosis signal-regulated kinase 1 (ASK1) is a serine/threonine kinase, MAP3K family member, which induces apoptosis through the activation of JNK and p38. ASK1 has been implicated in the pathology of neurodegenerative and oxidative stress-related diseases.

Apoptosis signal-regulated kinase 1 (ASK1) is a ubiquitously expressed serine/threonine kinase which induces apoptosis in a wide variety of cell types. ASK1 is a member of the MAP3K family and is therefore part of the MAPK signaling pathway. The kinase activates JNK and p38 through phosphorylation of SEK1/MKK7 and MKK3/MKK6 respectively, which induces mitochondria-dependent caspase activation and subsequent apoptosis. ASK1 is activated following auto-phosphorylation of Thr845 (mouse)/Thr838 (human) in the activation loop of its kinase domain, whilst Raf1 and the 14.3.3 proteins interact with ASK1 to suppress its proapoptotic activity.

ASK1-induced apoptosis can be activated by a number of stimuli, including: reactive oxygen species (ROS) such as hydrogen peroxide; death receptor ligands e.g. TNF-α and Fas ligand; endoplasmic reticulum (ER) stress; and lipopolysaccharide, GPCR and Ca2+ signaling. ASK1 has been implicated in the pathology of oxidative stress-related disease states such as ischemia-reperfusion injury and hematopoietic diseases, as well as in the role of ER stress in the pathogenesis of neurodegenerative diseases such as Alzheimer's, Huntington's and Parkinson's disease.
