    Protein Disulfide Isomerase (PDI, EC 5.4.3.1) is a multifunctional redox protein which acts as a regulator of disulfide bond formation and rearrangement (isomerization). In doing so it functions as both an enzyme and as a molecular chaperone.

    PDI, the first catalyst of protein folding to be characterized, is a member of the PDI family of proteins which are mainly localized to the endoplasmic reticulum (ER), are upregulated in response to ER stress and have an important role in protein homeostasis.

    This soluble 55 kDa protein consists of four thioredoxin-like domains arranged in a U-shape conformation with a C-terminal tail. Cysteine residues of a -CGHC- motif within the active sites of PDI react with thiol (R-SH) groups of substrate proteins to form disulfide bonds in a thiol-disulfide interchange.

    Alterations in cellular levels of PDI are associated with a range of diseases, for example, PDI upregulation has been shown to promote cancer cell survival in some types of breast cancer and thrombosis formation in cardiovascular disease. It may also have a protective effect in neurodegenerative disorders where it has been suggested to be a defence mechanism against protein aggregation.
