Submit ReviewSJ 10542 is a potent and selective JAK2/3 Degrader (PROTAC®) (DC50 values are 11 nM and 14 nM; Dmax = 92% and 90% for JAK3 and JAK2, respectively); uses a phenyl glutarimide ligand as the cereblon recruiter. Degrades JAK2 in ex vivo cell lines and patient derived xenografts (DC50= <120 nM) and displays high selectivity over GSPT1 and other members of the JAK family.
