Biological Activity

ProtiFect STAR™ Protein Transfection Reagent is a transfection reagent capable of delivering a wide range of proteins with varying molecular weights (up to 240 kDa) and isoelectric points to the cytoplasm, including Cas9 ribonucleoproteins and BSA-FITC. ProtiFect STAR™ is a gemini amphiphile, which bypasses endocytic pathways. Can be used in vivo and in vitro in a variety of cell types including primary cells, T-cells, B-cells and stem cells. This compound is supplied pre-dissolved in ethanol (10 mg/mL).

Scientific Data

Application of ProtiFect STAR™ Protein Transfection Reagent. Intracellular delivery of negatively charged green fluorescence protein (GFP), superoxide dismutase (SOD), ovalbumin (OVA), and positively charged lysozyme with a commercially available protein transfection reagent and ProtiFect STAR™. Proteins and ProtiFect STAR™ were at 8 μg/mL (n = 3). Scale = 25 μm.

Application of ProtiFect STAR™ Protein Transfection Reagent. Intracellular delivery of CRISPR/Cas9 ribonucleoprotein complexes with ProtiFect STAR™ in 293T cells targeting adeno-associated virus integration site 1 (AAVS1), leading to an indel frequency of 37.8%.

Application of ProtiFect STAR™ Protein Transfection Reagent. Intracellular delivery of CRISPR/Cas9 ribonucleoprotein complexes with ProtiFect STAR™ in SW-480 cells targeting KRAS locus. CMAX was used as a positive control. Sanger sequence (lower panel) performed by T-A cloning at KRAS locus was obtained from SW-480 cells. Three sequences of clones with mutations were represented. Cas9 protein, sgRNA, and ProtiFect STAR™ were 2, 1, and 4 μg/mL, respectively.
