Submit ReviewCoraFluor™ 2, amine reactive is a terbium-based time-resolved Förster resonance energy transfer (TR-FRET) donor for TR-FRET assay development, such as target engagement and ternary complex assays. CoraFluor™ 2 is designed with a PFP ester for reactivity with primary amines, making it suitable for labeling biomolecules, including antibodies, nanobodies, proteins and peptides. CoraFluor™ 2 is cell permeable and can be used in live cell intracellular TR-FRET assays. It is exceptionally bright, providing unmatched assay sensitivity and stability in biological media over other commercially available TR-FRET donors. CoraFluor™ 2 displays red shifted excitation and is excitable by both 365 nm and 405 nm lasers, and is compatible with common acceptor dyes such as FAM/FITC, AF488, GFP and mCherry, enabling seamless assay integration in high throughput screening applications.

CoraFluor™ 2 stability profile. Stability profiling of CoraFluor™ 2 and a commercially available TR-FRET donor (5 nM) after 7 days of exposure to 50 mM PBS, acetic acid (pH 3), sodium acetate (pH 5), MES (pH 6) or Tris buffer (pH 7.5) supplemented with various additives (DTPA, Ca2+, Mg2+, DTT, TCEP = 1 mM; Mn2+, Cu2+ = 0.5 mM).
Live cell TR-FRET assay. Cellular dose-dependent inhibition of HDAC1 via CoraFluor™ 2 labeled HaloTag® in HEK293T cells by various HDAC inhibitors (treatment for 4 h at 37 °C, 1 μM SAHA-NCT). HaloTag is a trademark of Promega Corporation.
