Casein kinase I (CK1) is a monomeric serine-threonine protein kinase with 7 isoforms: α, β, γ1, γ2, γ3, δ and ε. CK1 is involved in a number of cellular processes including DNA repair, cell division, nuclear localization and membrane transport.

CK1 isoforms also have key roles in the developmentally important Wnt and Hedgehog (Hh) signaling pathways. When Wnt is not bound to its receptor, CK1α is thought to phosphorylate β-catenin as part of its degradation process. Upon binding of Wnt to its receptor (Frizzled) CK1ε phosphorylates the intracellular signaling protein, Dishevelled and CK1γ directly phosphorylates the LRP5/6 co-receptor. These actions of CK1 facilitate signal transduction to the transcription factor, TCF, which mediates transcription of Wnt regulated genes. Similarly, CK1α plays a role in regulating Hh signaling by phosphorylating the signaling effector Ci-155 in the Hh unbound state, and then when Hh binds to Patched, CK1α phosphorylates the 7-TM co-receptor, Smoothened.
